Sulfadoxine has a structure similar to para-aminobenzoic acid(PABA) and blocks the conversion of PABA to the co-enzyme dihydrofolic acid , a reduced form of folic acid thtrathdrofolic acid is a co-enzyme that is important in the synthesis of pyrimethamine and purines which are essential in nucleic acid synthesis.
